Instead of questioning the culprit directly, you have to ramble about/fiddle with some unrelated thing in the room and know just when to pull out the slightly relevant "gotcha" question.
LA Noir works like that, but that's a video game.
I think the way to do it, is to have Columbo be an NPC or special player role and everyone else to be the web of relationships. The Columbo player had the npcs reveal more about who they are, and what they want.
It is important to have an established backstory to unfold, and the chance for the evil doer to get away with the crime, or else it'll just be a one-off that won't hold up over multiple plays (ahhh disposable games, my least favorite).
