Variation #1 & 2 are both really good! If I have to pick, #1 is best. I agree that the 45 degree angle variation makes it look like no brains are allowed.
I do like the diagonal line, though, because meds are often taken to make brains stop doing their thing. But I think I rate #1 highest, then #3, then #2 - the vertical line covers the hemisphere gap and makes it less obvious it's a brain, and the no brain connotation isn't for everyone.
